The 19-year-old forward played 58 matches for Madrid's first and second teams and registered 32 goals. He was also the top scorer at the UEFA European under-19 Championship in 2015, finishing the tournament with three goals.
Mayoral will join his new teammates for training at their camp at Bad Ragaz, Switzerland on Saturday.
"Wolfsburg are a big name in German football and in the Bundesliga," he said on his move, via Real's official website. "I'm very happy and excited about the new chapter which lies ahead. We want to qualify for the UEFA Champions League again. The team certainly has the potential to do that. From the first moment, I had a great feeling at Wolfsburg.
"I've had nine unforgettable seasons at Real Madrid. Right now I'm feeling a lot of emotions, but if I had to sum them all up, I'd give a heartfelt thanks for everything that the club has offered me and how it's helped me over the years. I'm going to try to perform to my best to come back here."
